خلاصـــة
تم تحضير مادة الأوكراتوكسين المرقمة بنظير اليود-125 باستخدام الكلورامين-ت كعامل مؤكسد وتمت دراسة العوامل المختلفة التى تؤثر على ناتج التفاعل مثل زمن التفاعل وتركيز كل من العامل المؤكسد والمادة المتفاعلة وأيضاً تأثير الأس الهيدروجينى. وقد استخدمت طرق الفصل الكروماتوجرافية مثل كروماتوجرافيا الطبقة الرقيقة وسيفادكس G-25 .
وقد تم إعطاء المادة المرقمة عن طريق الفم لفئران التجارب واختبار توزيعها فى أعضاء الجسم المختلفة وقد وجد أعلى تركيز لمادة الأوكراتوكسين المرقمة فى الكلى والكبد والدم والجهاز الهضمى مع عدم تكسير المادة المرقمة وثباتها داخل جسم حيوان التجارب.
ABSTRACT
Ochratoxin was successfully labelled
with 125I in presence of chloramine-T
(Ch-T) as an oxidizing agent. The effects of different reaction parameters
such as reaction time, oxidizing agent concentration, pH and concentration
of ochratoxin have been investigated. Separation and identification of the
labelled products were achieved by means of thin layer chromatography
(TLC) and gel chromatography using sephadex G-25 column. The labelled
product was orally administrated to rats and the accumulation of
radioactivity in the body at definite time intervals was examined. The
accumulation of the radioactivity was pronounced at the kidney, the liver,
the blood and digestive intestinal tract. The produced 125I-ochratoxin
was found stable in-vivo against biological decomposition.
